B/T Sure™ Biological Indicators (Thermo Scientific)

These autoclave controls are used as a biological indicator to monitor steam-under-pressure sterilization cycles. The population and D values of these indicators will vary depending on the lot made. The standard population value are 105 spores with a value of anywhere between 1.9 and 2.2 x 105. The D value is also typically in the range of 1.9 to 2.2.

Consists of bacterial spores of Bacillus stearothermophilus inoculated onto a paper carrier that is placed into a thermoplastic vial that serves as a culture tube. A small glass ampule containing sterile culture medium and color indicator is also contained in the vial. Upon completion of a sterilization cycle (121° to 135°C.), allow the B/T Sure to cool at least 10 minutes, then activate by compressing the plastic vial to break the glass ampule. This will allow the growth media to come in contact with the spore strip. After incubation at 55° to 60°C., the biological indicators are examined along with parallel controls not subject to the sterilization process. If growth in exposed indicators is visually observed, it must be assumed that the sterilization cycle was inadequate. Acid production during growth changes the color of the medium in the ampule from purple to yellow, facilitating detection of growth.
